Im Jahr 1984 wurde die frühere <a href="Typusart" class="mw-redirect" title="Typusart">Typusart</a> der Gattung, <i>M. mazei</i>, in die Gattung <i><a href="Methanosarcina" title="Methanosarcina">Methanosarcina</a></i> reklassifiziert und gleichzeitig <i><b>M. vannielii</b></i> als neue Typusart festgelegt.<sup id="cite_ref-Mah1984_6-0" class="reference"><a href="#cite_note-Mah1984-6"><span class="cite-bracket">[</span>6<span class="cite-bracket">]</span></a></sup>
Im Jahr 2002 erfolgte eine weitere Aufspaltung der Gattung durch Abspaltung hyperthermophiler Arten in die Gattung <i><a href="Methanocaldococcus" title="Methanocaldococcus">Methanocaldococcus</a></i>.<sup id="cite_ref-9" class="reference"><a href="#cite_note-9"><span class="cite-bracket">[</span>A. 1<span class="cite-bracket">]</span></a></sup>
Die in der Gattung verbliebenen Spezies sind mit Ausnahme der <a href="Thermophil" class="mw-redirect" title="Thermophil">thermophilen</a> Art <i>M. thermolithotrophicus</i> alle <a href="Mesophil" class="mw-redirect" title="Mesophil">mesophil</a>, d.&nbsp;h. sie bevorzugen gemäßigte Temperaturen.

<div class="mw-heading mw-heading2"><h2 id="Beschreibung">Beschreibung</h2></div>
<p>Die Zellen der Typusart <i>M. vannielii</i> sind annähernd <a href="Kokken" title="Kokken">kokkoid</a> und variieren in einer Kultur von 0,5 bis 4,0&nbsp;<a href="Meter#µm" title="Meter">µm</a>, gewöhnlich zwischen 1 und 2&nbsp;µm.
Sie sind oft leicht elliptisch und treten häufig paarweise auf. Manchmal ist ein Mitglied eines solchen Paares viel kleiner als das andere und ähnelt daher einer <a href="Knospung" title="Knospung">Knospe</a> auf einer <a href="Hefezelle" class="mw-redirect" title="Hefezelle">Hefezelle</a>. Wenn die Zellen aus tiefen Agar-Kolonien entnommen werden, haben sie oft zunächst sehr verzerrte Formen, nehmen aber bald wieder eine kugelförmige Gestalt an.
Der Organismus ist aktiv beweglich, die Zellen beschreiben einzeln oder paarweise eine spiralförmige Bahn.
Die <a href="Zellwand" title="Zellwand">Zellwand</a> hat sich als sehr empfindlich gezeigt. Mikroskopische Untersuchungen müssen beispielsweise an Nasspräparaten durchgeführt werden, da die Zellen bei der üblichen Herstellung von getrockneten, gefärbten Ausstrichen zerfallen und dann fast nicht mehr zu erkennen sind. Außerdem werden die Zellen auch durch die Zugabe von <a href="Methylcellulose" title="Methylcellulose">Methylcellulose</a> (üblicherweise zur Verlangsamung der Motilität) zerstört.
Tiefe Agar-Kolonien sind linsenförmig und hellbraun und haben einen Durchmesser von 0,5 bis 1,0&nbsp;<a href="Millimeter" class="mw-redirect" title="Millimeter">mm</a>, unter dem Mikroskop erscheint die Oberfläche der Kolonie körnig.<sup id="cite_ref-Stadtmann1951_10-0" class="reference"><a href="#cite_note-Stadtmann1951-10"><span class="cite-bracket">[</span>9<span class="cite-bracket">]</span></a></sup>

<p><a href="Formiat" class="mw-redirect" title="Formiat">Formiat</a> begünstigt ein gutes Wachstum <i>M. vannielii</i>; maximales Wachstum wurde bei einer Formiatkonzentration von 1,5&nbsp;% erreicht, bei 2 bis 3&nbsp;% verlangsamt sich das Wachstum und kommt bei 5&nbsp;% zum Erliegen.
Der Ersatz von 0,05&nbsp;% NH<sub>4</sub>Cl (<a href="Ammoniumchlorid" title="Ammoniumchlorid">Ammoniumchlorid</a>) durch 0,05 bis 0,1&nbsp;% (NH<sub>4</sub>)<sub>2</sub>SO<sub>4</sub> (<a href="Ammoniumsulfat" title="Ammoniumsulfat">Ammoniumsulfat</a>) hatte einen leicht positiven Effekt auf das Wachstum, höhere Konzentrationen von Ammoniumsulfat wirkten hemmend.
Der <a href="Ph-Wert" class="mw-redirect" title="Ph-Wert">pH-Bereich</a> für das Wachstum liegt zwischen 7,4 und 9,2, mit einem Optimum bei pH&nbsp;8. Während der Fermentation von Natriumformiat steigt der pH-Wert tendenziell schnell an, da dadurch die relativ starke Säure <a href="Ameisens%C3%A4ure" title="Ameisensäure">Ameisensäure</a> in <a href="Methan" title="Methan">Methan</a> und die schwache Säure <a href="Kohlendioxid" class="mw-redirect" title="Kohlendioxid">Kohlendioxid</a> (Kohlensäure) umgewandelt wird.<sup id="cite_ref-Stadtmann1951_10-1" class="reference"><a href="#cite_note-Stadtmann1951-10"><span class="cite-bracket">[</span>9<span class="cite-bracket">]</span></a></sup>
